Biomolecular simulations can be used to address some of the world's most pressing healthproblems. One example is the development of new antibiotics against resistant bacteria. Every year, morethan one milion people die because conventional antibiotics have become ineffective due to the developmentof bacterial resistance mechanisms. Starting from the essential medicine clarithromycin, it is showwn how tofight the evolutionary adaptation of bacteria with a computer-driven adaptation ofthe antibiotic. inparticulatsimulations of the solubility, membrane permeability, and binding affinity towards the bacterial ribosome ofa series of clarithromycin derivatives are used to overcome bacterial resistance mechanisms. The resultingnew antibiotic is on average 5.8 times more potent than clarithromycin and shows activity against the threetop priority targets of the World Health Organization.

Gerhard Koenig博士2010年毕业于奥地利维也纳大学并取得博士学位,2010-2018年先后在美国国立卫生研究院(NIH)、马克斯·普朗克研究所、罗格斯大学,新泽西州立大学担任访问学者;2018-2020年作为博士后研究员在苏黎世联邦理工学院与Sereina Riniker教授合作,开发了多肽药物的膜渗透性、λ包络分布采样方法。2020-2024年,作为高级访问学者加入朴茨茅斯大学,如今在Recursion Pharmaceuticals担任副主任,并致力于基于机器学习、分子力学和量子力学的抗病毒和抗癌药物的计算药物发现工作。
发表了多篇论文,其中作为第一作者21篇, 作为通讯作者:16篇, h-index(Google Scholar):23, 论文占比80.0% Q1期刊。
